چکیده

The densities of positively charged, colloidal iron hydroxide particles binding to the surfaces of glutaraldehyde-fixed cultured Ehrlich ascites tumour cells, were determined on cells maintained in suspension and monolayer cultures. In both cases, there was a higher density of particles bound to the surfaces of microvilli than to the spaces between them. Prior treatment of the cells with ribonuclease did not significantly alter the proportions of particles binding to the 2 different surface regions, indicating that the ribonuclease-susceptible binding sites are distributed with similar mean densities over the microvilli and the regions between them. Prior treatment of the cells with neuraminidase reduced the densities of particles binding to microvilli to those of the intermicrovillous spaces, indicating that the original higher density over the microvilli is due to an increased number of sialic acid residues at their surfaces. The findings are discussed in relationship to observed changes in the surface density of microvilli when these cells convert from suspension to monolayer cultures.

ANIONIC SITES OF HUMAN ERYTHROCYTE MEMBRANES I. Effects of Trypsin, Phospholipase C, and pH on the Topography of Bound Positively Charged Colloidal Particles

The effects of pH, trypsin, and phospholipase C on the topographic distribution of acidic anionic residues on human erythrocytes was investigated using colloidal iron hydroxide labeling of mounted, fixed ghost membranes . After glutaraldehyde fixation at pH 6 .5-7 .5, the positively charged colloidal particles were bound to the membranes in small randomly distributed clusters. The clusters of a...

Expression of anionic sites on tumour cells at different stages of tissue invasion in vivo: a comparative study by X-ray microanalysis.

Quantification of colloidal iron hydroxide (CIH) labelling by X-ray microanalysis was used to investigate anionic sites at the surface of Ehrlich carcinoma cells from different locations in the mouse host. Individual tumour cells from peritoneal ascites suspensions (pre-invasion stage) varied up to threefold in their ability to bind CIH and a similar degree of intra-tumour heterogeneity was obs...

P-120: The Impact of Progesterone and Sildenafil on Endometrial Ultrastructure in Superovulated Mice

Background: Implantation is proceeded by a process referred as adhesion. Adhesion requires maturation of endometrium and blastocyst to confer adherence ability to them. During IVF cycles, endometrial maturation is facilitated by progesterone administrating. Considering vasodilatory effect of Sildenafild, in genital organs, it appears that Sildenafil may have similar effect as progestrone. The a...

Antitumor Activity of Prosopis glandulosa Torr. on Ehrlich Ascites Carcinoma (EAC) Tumor Bearing Mice

The antitumor activity of ethanol extract of Prosopis glandulosa Torr. (EPG) was evaluated against Ehrlich ascites carcinoma (EAC) tumor model in Swiss albino mice on dose dependent manner. The activity was assessed using survival time, average increase in body weight, hematological parameters and solid tumor volume. Oral administration of EPG at the dose of 100, 200 and 400 mg/Kg, significantl...
